Still no compression!

Unable to cure misfire after changing the usual suspects, did a compression test and got the following...
No.1 - 60psi
No.2 - 60psi
No.3 - 0psi
No.4 - 30psi
So dropped the engine to check heads, a bit sooty around the top suggesting a leak. Had the heads inspected and cleaned, new exhaust valve guides bushed as they were worn and valves reseated. Fitted new piston rings, barrels still have honing marks on and nothing else wrong really.
Fitted head on one side before running out of light last night, now...
No.3 - 20psi
No.4 - 30psi.
Should I be doing a compression test by turning the engine over? Are the readings lower by winding the engine over by hand?

Any engine builders have any thoughts?

you do need to have the engine turned by the starter motor you wont get any good readings by hand cranking, all 4 spark plugs should be removed & carb needs to be fully opened ht dissy & fuel isolated off (remove - on coil & wire to solenoid on carb + hold make sure the carb is fully opened to let more air in)

Easier with two people one to crank the engine whilst you sit by the engine bay or you can make a temporary connection to starter motor you need a decent piece or wire crimped one end plug onto starter motor solenoid thinner wire to starter needs to be long enough to reach round to your battery just touch it on the positive terminal to crank engine when ready take your reading & move to next cylinder & repeat.
